Effect of Overexpression of Constitutively Active PKCα on Rat Lacrimal Gland Protein Secretion
نویسندگان
چکیده
PURPOSE. The lacrimal gland secretes water, electrolytes, and protein into the tear film. Decreased secretion from the lacrimal gland can lead to dry eye syndromes with deleterious effects on vision. Protein kinase C (PKC)plays a major role in cholinergicand 1-adrenergic–induced protein secretion from the lacrimal gland. This study was undertaken to determine whether activation of PKC alone would induce lacrimal gland protein secretion by examining the effects of overexpression of constitutively active PKC .

Effect of overexpression of constitutively active PKCalpha on rat lacrimal gland protein secretion.
PURPOSE The lacrimal gland secretes water, electrolytes, and protein into the tear film. Decreased secretion from the lacrimal gland can lead to dry eye syndromes with deleterious effects on vision. Protein kinase C (PKC)-alpha plays a major role in cholinergic- and alpha1-adrenergic-induced protein secretion from the lacrimal gland. This study was undertaken to determine whether activation of ...
متن کاملCholinergic agonists activate P2X7 receptors to stimulate protein secretion by the rat lacrimal gland.
PURPOSE To determine the interaction of M3 muscarinic receptors (M3AChR) and P2X(7) receptors to increase intracellular [Ca2+] ([Ca2+]i) and stimulate protein secretion in rat lacrimal gland cells. METHODS Exorbital lacrimal glands from male Sprague-Dawley rats were divided into pieces or digested with collagenase to form acinar clumps. [Ca2+]i was measured using an imaging system in acini in...
متن کاملAltered morphology and function of the lacrimal functional unit in protein kinase C{alpha} knockout mice.
PURPOSE Protein kinase C (PKC) α plays a major role in the parasympathetic neural stimulation of lacrimal gland (LG) secretion. It also has been reported to have antiapoptotic properties and to promote cell survival. Therefore, the hypothesis for the present study was that PKCα knockout ((-/-)) mice have impaired ocular surface-lacrimal gland signaling, rendering them susceptible to desiccating...
متن کاملCharacterization of P2X7 purinergic receptors and their function in rat lacrimal gland.
PURPOSE To characterize the effects of P2X(7) purinergic receptors on lacrimal gland function. METHODS P2X(7) purinergic receptors were identified by RT-PCR, Western blot analysis, and immunofluorescence techniques. Rat lacrimal gland acini were isolated by collagenase digestion. Acini were incubated with the fluorescent indicator molecule fura 2, and [Ca(2+)](i) was measured by a fluorescenc...
متن کاملRoles of caspase 1 and extracellular signal-regulated kinase in inflammation-induced inhibition of lacrimal gland protein secretion.
PURPOSE The purpose of the present study was to investigate the roles of caspase 1 and extracellular signal-regulated kinase (ERK) in inflammation-induced inhibition of lacrimal gland secretion. METHODS Lacrimal gland inflammation was induced by injection of lipopolysaccharide (LPS; to study the role of caspase 1) or IL-1beta (to study the role of ERK). Lacrimal gland protein secretion was me...
